Course Description: A 1-day course involving classroom and laboratory sections for DNA library construction for sequencing on the Ion Torrent™ systems.

Attendees will gain hands-on experience in constructing a DNA fragment library using control genomic DNA, and learn about different methods for shearing and size-selection, library quality assessment, amplification, quantification and troubleshooting.

Ideally, individuals have already participated in the live, online System and Library Training, although this is not mandatory.

If this is the first training after instrument purchase, it is mandatory to have your instrument fully operational; being able to use the instrument right after the training guarantees the best use of your time.

(Please note, we will NOT be able to use customer samples for library construction during this course.) Information and strategies for different sequencing applications and how these relate to library construction and preparation of genomic samples will also be discussed.

Course topics include an overview of instrument operation and hardware, library preparation, Ion Sphere™ particles preparation and deposition, quality control runs, troubleshooting, and data analysis.

An overview of the chemistry used in the Ion Torrent™ Personal Genome Machine™ will be provided, which is critical for understanding the system and troubleshooting.
